人脑胶质瘤细胞对诱导分化药物敏感性的研究
The Sensibility of Human Glioma Cell to Seven Differentiation Inducing Agents.
【摘要】 目的 优选人脑胶质瘤细胞诱导分化剂。方法 应用MTT法、形态学、GFAP免疫荧光等方法鉴定环已亚甲基二乙酰胺、二甲基甲酰胺、丁酸钠、二丁酰杯AMP、苯乙酸钠、苯丁酸钠、亚砷酸钠等7种诱导剂对人脑胶质瘤细胞系SHG-44的诱导分化效果。结果 亚砷酸钠、苯乙酸钠抑制肿瘤细胞增殖效果较差,苯丁酸钠、二丁酰环AMP、丁酸钠在1-2mM即可明显诱导人脑胶质瘤细胞多分化,表现为增殖抑制,胞突增长,GFAP表达升高现象。结论 人脑胶质瘤细胞对诱导分化剂苯丁酸钠、二丁酰环AMP和丁酸钠较为敏感,值得深入研究。
【Abstract】 AIM To study the sensibility of human glioma cells to 7 differentiation inducing agents, and screen out those most effective for glioma therapy. METHODS Human glioma cell line SHG- 44 is induced to differentiation with Hexamethelene bisacidamide (HMBA) , Dimethylformamide (DMF) , Sodium butyrate (BA) , Dibutyric cycle AMP(dBcAMP) , Sodium phenylacetate(SPA) , Sodium phenyIbutyrate (SPB), Asenic trioxide, and differentiation level was studied by MTT test, morphorlogy, GFAP immunoflurecency. RESULTS To glioma cells, asenic trioxide and SPA have little effect. SPB, dBcAMP, BA are better agents, and in concentration of 1-2 mmol/L, they can induce obvious growth attest, extension of cell process, GFAP expression increasing. CONCLUSION Human glioma cells are more sensitive to dii’ferentiation inducing agents SPB, dBcAMP and BA.
