表面抗原P43基因在异基因造血干细胞移植患者弓形虫感染诊断中的应用(英文)
Amplification of Surface Antigen P43 Gene and Its Application in Detection of Toxoplasma Gondii in Allogeneic Hematopoietic Stem Cell Transplantation
【摘要】 目的 建立快速、特异、敏感诊断造血干细胞移植受者的弓形虫的感染情况的DNA检测方法 ,并讨论它的临床意义。方法 利用ELISA和PCR方法对 30例异基因造血干细胞移植受者进行检测。结果 检测异基因造血干细胞移植受者 30例 ,PCR和ELISA方法检测弓形虫表面抗原P43基因和其抗原 ,阳性各为 5例及 3例 ;其阳性率分别为 16 .6 %和 13.3%。作为阴性对照的 2 0名正常对照组同时进行弓形虫的检测 ,其结果均为阴性。结论 体外扩增弓形虫P43基因的方法具有准确、快速、特异和敏感的优点 ,因为其适用于临床的各种不同标本 ,PCR被认为是对弓形虫感染作出诊断很有价值的一种手段。
【Abstract】 Objective To establish a rapid, specific and sensitive diagnostic technique for the human Toxoplasma gondii infection in the recipients with allogeneic hematopoietic stem cell transplantation and discuss its clinical significance. Methods 30 patients undergoing allogeneic hematopoietic stem cell transplantation were detected by using ELISA and PCR. Results Among 30 recipients undergiong allogeneic hematopoietic stem cell transplantation , 3 were positive for Toxoplasma gondii antigen and 5 for surface antigen p43 gene with the positive rate being 13.3% and 16.67% respectively. 20 healthy people (negative for anti-Tox antibody) were also tested by using ELISA and PCR. Conclusion PCR is an accurate, relatively rapid, sensitive and specific method for detecting P43 gene of Toxoplasma gondii. Because PCR can be applied to a variety of different clinical samples, it can be considered as a valuable additional tool for identification of Toxoplasma gondii infections.
