褐藻岩藻糖胶对小鼠脂质过氧化的影响
EFFECTS OF FUCOIDAN EXTRACTED FROM BROWN SEA WEED ON LIPID PEROXIDATION IN MICE
【摘要】 目的 : 探讨褐藻海带中岩藻糖胶 (FD)对抗机体组织脂质过氧化的作用。方法 : 昆明种小鼠分六组 ,分别经口给予 1 0 ,5 0 ,1 5 0及 3 0 0 mg/(kg·d)剂量 FD,连续 7d,尾静脉注入四氧嘧啶 70 mg/kg。 4d后 ,测定血清、肝及脾组织过氧化脂质 (LPO)含量 ;另取小鼠 ,以四氧嘧啶造成糖尿病模型 ,分组经口给予不同剂量的 FD7d,测定 LPO;取正常小鼠肝及脾脏 ,制成匀浆 ,分别加入 0 .0 5 % ,0 .2 5 % ,0 .75 %及 1 .5 % FD溶液各 0 .2 ml,测 LPO;另取匀浆 ,同时加入半胱氨酸(Cys) /Fe SO4 及不同浓度 FD溶液 ,测 LPO。结果 : 预先给予 FD的各组 ,注射四氧嘧啶后 ,血清、肝及脾中 LPO明显低于阳性对照组 ,以 5 0 mg/kg作用较强 ,分别降低 3 2 .7% (P<0 .0 0 1 ) ,2 2 .7%及 2 0 .6 % (P<0 .0 1 ) ;糖尿病小鼠给予 5 0 mg/kg FD,血清、肝及脾组织 LPO分别降低3 4 .1 % ,2 9.3 %及 3 0 .3 % ;小鼠组织匀浆中加入 FD,或同时加入 FD及 Cys/Fe SO4 ,各组 LPO无统计学差异。结论 : FD对四氧嘧啶损伤引起的 LPO升高有一定预防作用 ,对降低四氧嘧啶糖尿病小鼠 LPO水平有较好作用 ,但对正常小鼠肝、脾组织匀浆脂质体外自氧化及受 Cys/Fe SO4 激发过氧化均无明显抑制作用
【Abstract】 Objective: To investigate the effects of fucoidan on the production of lipid peroxide (LPO) in mice. Methods: (1) Fucoidan was orally administered to mice at doses of 10, 50, 150, and 300 mg/kg·d for 7 days. Then all mice were injected intravenously with 70 mg/kg alloxan, 4 days later, LPO content in serum, liver, and spleen was determined. (2) Different doses of fucoidan were orally administered to alloxan-induced diabetic mice for 7 days, and LPO level was assayed. (3) In vitro, 0.05%, 0 25%, 0.75%, and 1.5% fucoidan solution only or with Cys/FeSO 4 together were added into liver or spleen homogenates, and LPO was determined. Results: Those groups administered with fucoidan prior to alloxan injection had remarkable low LPO values, as compared with experimental control. Fucoidan (po 50 mg/kg·d) prevented the increase of LPO in serum, liver and spleen of diabetic mice by 32 7% (P<0 001), 22.7% and 20.0% (P<0.01), respectively. It also obviously reduced LPO levels in serum, liver and spleen of diabetic mice by 34.1%, 29.3%, and 30.3%, respectively. No statistical differences were found in LPO levels between liver or spleen homogenates added with fucoidan only or with Cys/FeSO 4 together. Conclusion: Fucoidan (po 50 mg/kg·d) can prevent the increase of LPO in serum, liver and spleen of diabetic mice obviously. However, no inhibition effect was found on both spontaneous lipid peroxidation of homogenates and that induced by Cys/FeSO 4 in vitro.
