【摘要】 目的 :建立正气扬质量控制方法。方法 :采用薄层色谱法 (TLC)以黄芪甲苷和刺五加药材为对照品 ,分别对其主药黄芪和刺五加进行定性鉴别 ;运用改良苯酚硫酸法 ,对其主要成分多糖进行含量测定。结果 :样品TLC色谱图中均能显示黄芪及刺五加药材的特征斑点 ;样品经苯酚硫酸显色后 ,于 488nm测得吸光度与葡萄糖量呈良好线性关系 ,r =0 .9994,加样回收率 =97.94% ,RSD =2 .3 3 % (n =6) ,测得 6批样品多糖含量相当于葡萄糖 2 0~ 40mg·mL 1 。结论 :TLC法可用于本品定性鉴别 ,改良苯酚硫酸法可用于其多糖含量测定

【Abstract】 Objective:To establish a method for quality evaluation of Zhengqiyang mixture mainly composed of Radix Astragali and Radix Acanthopanacis Senticosi . Methods:The preparations of Radix Astragali and Radix Acanthopanacis Senticosi were selected as the control samples. The thin layer chromatography (TLC) was employed to characterize the control as well as the main components of Zhengqiyang mixture. The modified phenol sulfate method was used to quantitate polysaccharides in the preparations in conjunction with spectrophotometry. Results: Spots in the diagram of TLC were clear and the spot of astragaloside A showed purple with Rf=0.58, T=20℃ and RH=67; Meanwhile, TLC performed to identify Radix Acanthopanacis Senticosi showing that the target spot was blue with Rf=0.78,T=25℃ and RH=70%. The absorptions of processed samples were tested at the wavelength of 488 nm by phenol sulfate. The average recovery was 97.94% with RSD of 2.33%, the measured contents of polysaccharides in 6 batches of preparations were equivalent to 20~40 mg·ml 1 of glucose. Conclusions: TCL can be applied for identifying the main components of the mixture, and the content of polysaccharides in the products can be measured by the modified phenol sulfate method.
