复方石韦片中生物碱的分离测定

【摘要】 目的:建立了复方石韦片中槐果碱和苦参碱的分离测定方法。方法:采用高效毛细管电泳法进行测定。运行电压27kV;运行缓冲液为0.2 mol·L-1 Tris-40 mmol·L-1磷酸二氢钠-5%异丙醇,磷酸调pH 5.75;进样方式:真空进样;进样时间:5s;检测波长:205nm。结果:槐果碱和苦参碱的线性范围分别为0.5~5.76μg·mL-1和0.66~13.2μg·mL-1,相关系数分别为0.9991和0.999 3,平均回收率分别为99.5%和99.3%,RSD分别小于4%和2%(n=3)。结论;本法简便、快速、重现性好。

【Abstract】 Objective: A new method was developed for the separation and assay of sophocarpine and matrine in compound Shiwei tablets. Method: The content of the alkaloids in tablets was determined using high performance capillary electrophoresis. The injection mode was hydrodynamic, the injection time was 5 s, the voltage applied was 27 kV,0. 2 mol L-1Tris -40 mmol ·L sodium dihydrogen phosphate -5% isopropanol was selected for the optimal running buffer, adjusted to pH 5.75 with 0. 1 mol·L-1 phosphate acid. The detection wavelength was 205 nm. Results:The linear ranges were 0. 5-5. 76μg·mL-1 for sophocarpine and 0. 66 - 13. 2μg·mL-1 for ma-trine with correlation coefficient at 0. 999 1 and 0. 999 3 respectively. The average recoveries of the method were 99. 5% and 99. 3% , and the RSDs of injection were less than 4% and 2% for sophocarpine and matrine respectively ( n = 3). Conclusion:This method was simple, rapid with good reproducibility.
