视网膜神经节细胞多种培养方法的实验研究

【摘要】 目的探讨视网膜神经节细胞(RGCs)的多种培养方法。方法(1)在涂有鼠尾胶原的器皿上通过调整培养液培养:①乳鼠混合及纯化的RGCs;②单层星型胶质细胞上种植纯化的乳鼠RGCs;③引产胎儿混合RGCs。(2)检验鼠尾胶原及中脑顶盖提取液(Te)对培养的乳鼠RGCs的影响。结果(1)混合培养的乳鼠及引产胎儿RGCs大多能存活2周以上;(2)纯化的乳鼠RGCs大多能存活48 h;(3)单层星型胶质细胞上种植的纯化乳鼠RGCs能存活4周以上;(4)鼠尾胶原及Te对培养的乳鼠RGCs的贴壁及生长有明显的促进作用。结论通过调整培养液及改善微环境能用多种方法培养RGCs。

【Abstract】 ObjectiveTo study different culture methods of retina ganglion cells (RGCs).Methods(1)To culture RGCs on utensils that smeared rat’s tail collagen with culture solution:①co-cultivation and pure culture of newborn rat’s RGCs;②culture purified RGCs of newborn rat on subculture monolayer astrocyte;③co-culture of induced labor fetus’s RGCs.(2)To determinate effect of Te and rat’s tail collagen on RGCs.Results(1)Most cocultivation of newborn rat’s RGCs and induced labor fetus’s RGCs could survive for more than 2 weeks;(2)Most of newborn rat’s RGCs could survive for 48 hours;(3)culture purified RGCs of newborn rat on subculture monolayer astrocyte of rat brain could survive for more than 4 weeks;(4)To smear rat’s tail collagen on cell culture utensils and using Te (retinal ganglion neuronotrophic facter,RGNTF) could benefit RGCs to survive.ConclusionRGCs could be cultured on rat’s tail collagen with culture solution of DMEM,N3,bovine serum and Te.
