一个新的猪肌肉组织EST的分离、定位与表达
Isolation, Localization and Expression of a Novel Expressed Sequence Tag from Pig Skeletal Muscular Tissue
【摘要】 利用mRNA差异显示技术 ,从猪骨骼肌组织中分离到一个新的表达序列标签 (expressedsequencetag,EST)ESThp9 1(其GenBank登录号 :BI5 96 2 6 2 ) ,其序列长 196bp ,经BLAST程序与GenBank中存在的序列比对后 ,发现与猪的所有序列无同源性 ,但与大鼠的U3A核内小RNA基因及小鼠的U3B .4核内小RNA基因同源性分别为 87% (93个碱基 )和 85 % (96个碱基 )。半定量RT PCR表明 ,此EST在猪的多数组织中均有表达。通过体细胞杂种板(somaticcellhybridpanel,SCHP)及辐射杂种板 (radiationhybridpanel,RH)分析 ,ESThp9 1被定位于猪的 12号染色体长臂 ,与微卫星标记S0 0 90连锁。根据同源性比对和物理定位结果 ,推测ESThp9 1为猪的U3基因家族中一员。
【Abstract】 A novel expressed sequence tag (ESThp9 1, GenBank accession number: BI596262) was isolated from pig skeletal muscular tissue by using the mRNA differential display technique. BLAST analysis revealed that the 196bp long EST (ESThp9 1) was not homologous to any of the known porcine genes in the database but similar to rat U3A small nuclear RNA (87% identity over 93 nucleotides) and mouse U3B.4 small nuclear RNA (85% identity over 96 nucleotides). Semi quantitative reverse transcription polymerase chain reaction indicated that EST9hp 1 was expressed in most of tissue of the pig. ESThp9 1 was physically mapped on sus scrofa chromosome 12q1.1 q1.5 and linked with microsatellite S0090 by using somatic cell hybrid panel and radiation hybrid panel analysis. According to the homologous information and result of physical mapping, ESThp9 1 was presumed to be one member of the porcine U 3 gene family.
