免疫金银染色法用于检测系统红斑狼疮患者血清抗核抗体和抗双链DNA抗体
Detection of ANA and A-dsDNA antibodies in the sera of patients with SLE by IGSS and Dot-IGSS
【摘要】 目的 探讨检测抗核抗体 (ANA)、抗双链DNA抗体 (A -dsDNA)对系统性红斑狼疮 (SLE)的诊断意义。方法 采用免疫金银染色法 (IGSS)和用斑点免疫金银染色法 (Dot-IGSS) ,检测SLE病人血清中ANA和A -dsD NA。结果 113例活动期SLE患者血清ANA阳性率 97.3% ,A -dsDNA阳性率 71.7% ;35例非活动期SLE患者血清ANA阳性率 74 .3% ,A -dsDNA阳性率 14 .3%。活动期ANA、A -dsDNA阳性率与非活动期比较均有显著性差异 (P <0 .0 1)。活动期ANA平均滴度明显高于非活动期 ,A -dsDNA阳性组肾损害明显高于A -dsDNA阴性者(P <0 .0 1)。结论 IGSS和Dot-IGSS检测ANA、A -dsDNA有高度特异性、敏感性和稳定性 ,较高滴度A -dsD NA提示有肾损的可能
【Abstract】 Objective To detect the anti-nuclear antibody (ANA) and anti-double stranded DNA(A-dsDNA) in the sera of patients with SLE. Methods The ANA and A-dsDNA antibodies in the sera of 148 patients with SLE were detected by using immunogold-silver staining (IGSS) and Dot-IGSS. Results The positive rates of ANA and A-dsDNA were 97.3% and 71.7% respectively in 113 active cases of SLE, and 74.3% and 14.3% respectively in 45 inactive cases. The difference in the positive rates of ANA and A-dsDNA was significant between the active and inactive cases (P<0.01), and the titers of ANA in active cases were higher than that in the inactive cases (P<0.01). The incidence of renal damage in patients with positive A-dsDNA was higher than that in patients with negative A-dsDNA. Conclusion The IGSS and Dot-IGSS are highly sensitive and reliable for detecting ANA and A-dsDNA in patients with SLE. And the higher titer of A-dsDNA suggest the possibility of renal damage.
