拟除虫菊酯对大鼠脑组织中白介素-1β的影响
The effect of pyrethroid on the expression of IL-1 β in rat brain
【摘要】 目的 探讨细胞因子与拟除虫菊酯中枢神经系统损害机制的关系。方法 用逆转录 聚合酶链式反应 (RT PCR)和免疫组织化学的方法观察了拟除虫菊酯对大鼠脑组织中白细胞介素 1beta(IL -1β)表达的影响。 结果 大鼠经氯菊酯一次大剂量和多次小剂量处理后 ,其皮层和海马的IL -1βmRNA水平有明显的升高 ,分别是对照组的 2 1、1 9和1 9、1 9倍 ;而一次大剂量氯菊酯处理后其蛋白水平的表达在海马尤其是CA3区显著升高 ,达到对照的 1 5倍 ,皮层则无明显变化 ;小剂量反复给药后皮层和海马的蛋白表达均明显上升。溴氰菊酯处理后IL -1β的mRNA也有升高趋势 ,而蛋白水平没有明显的改变。结论 拟除虫菊酯对IL -1β快速诱导可能是脑组织对损伤的一种保护反应。
【Abstract】 Objective To investigate the relationships between cytokine and the mechanism of central nervous system impaired by pyrethroid. Methods Permethrin(PM) was administered with daily doses of 400 mg/kg for one day and 200 mg/kg for 5 days; and Deltamethrin (DM) was administered with daily doses of 25 mg/kg for one day and 12.5 mg/kg for 5 days, respectively. The expression of IL 1β in rat brain was observed by RT PCR and immunohistochemistry. Results A significant increase in IL 1β mRNA in cortex and hippocampus was recorded after treatment with PM and the protein expression of IL 1β appeared to be increased in hippocampus only. Whereas the change of IL 1β mRNA and protein expression was mild after treatment with DM. Conclusion These results indicated that the pyrethroids induced IL 1β expression might be a protective response of brain against an impairment.
