当归、川芎嗪对家兔骨骼肌缺血再灌注损伤作用的研究
Reperfusion Injury Effect of Chinese Angelica and Ligustrazine on Skeletal Muscles of Rabbit
【摘要】 目的:观察当归和川芎嗪注射液对家兔骨骼肌缺血再灌注损伤的作用。方法:制备家兔右后肢缺血再灌注模型,缺血时间5h后,实验组分别由股动脉注射当归注射液2ml/kg(含原药材1g/ml)或川芎嗪注射液2ml/kg(20mg/ml),对照组则由股动脉注射等容生理盐水。再灌注1h,测定患肢静脉血中丙二醛(MDA)、肌酸磷酸激酶(CPK)、天冬氨酸转氨酶(AST)、乳酸脱氢酶(LDH);光镜下观察缺血组织的病理变化。结果:单纯应用当归或川芎嗪可使MDA含量降低,CPK、AST和LDH升高,可加重家兔缺血再灌注对骨骼肌组织的损伤。结论:当归、川芎嗪注射液可加重家兔缺血再灌注损伤。
【Abstract】 Objective:The study aimed at observing the reperfusion injury of Chinese Angelica and Ligustrazine to the skeletal muscles of rabbit. Methods:Twenty-four models of ischemic reperfusion of the rabbit hind leg were made. After 5 hour’s ischemia, solutions of Chinese Angelica〔2ml/kg (1g/ml)〕 and Ligustrazine〔2ml/kg (20mg/ml)〕 were infused into femoral arteries of the two groups of 16 experimental rabbits, respectively. The 8 control rabbits were infused with same amount of physical saline. After 1 hour’s reperfusion, the venous blood samples were taken and the levels of plasma malondialdehyde(MDA), creatine phosphokinase(CPK), aspartate aminotransferase (AST) and lactate dehydrogenase(LDH) were measured. Pathologic alterations of the ischemic tissue were observed with optic microscope. Results:Both Chinese Angelica and Ligustrazine decrease the levels of MDA. However they increase the levels of CPK, AST and LDH and aggravate the reperfusion injury of skeletal muscles of rabbits. Conclusion:Parenteral solutions of both Chinese Angelica and Ligustrazine can aggravate the reperfusion injury of skeletal muscles of rabbits.
