【摘要】 用人抑制素α亚基 (1 2 6 )Gly·Tyr与KLH连接作免疫原 2 0mg ,免疫本地良种母鸡 ,旨在获得大量抗抑制素抗体。用间接ELISA法测定表明 :经免疫 ,母鸡的血清、卵黄中有大量的高效价抗体。综合运用pH值、非球蛋白沉淀法和球蛋白沉淀法 ,制得经纯化的抑制素抗体 2 5 6 40mL。采用阻断ELISA法和大鼠子宫、卵巢增重试验 ,证实所获得的抑制素抗体具有高度的特异性。可以用鸡做生物反应器大量高效生产抗抑制素抗体更多还原

【Abstract】 Laying hens were actively immunized with synthetic human inhibin peptide fragment (hIHN(1 26)Tyr.Gly) conjugated to either keyhole limpet hemocyanin(KLH,n=8)or saline alone(n=12).The aim is to attain large amount of anti inhibin eIgY.We studied the antibody in serum egg.Six hens have produced the needed antibody.Purified anti inhibin eIgY was made in two steps.Inhibit ELISA analysis demonstrated that the antibody isolated from egg yolk had high speciality.Indirect ELISA analysis showed that it also had high affimity,and OD 450 of eIgY and purified eIgY were equal to that of sIgY.Three days continuous injection of eIgY could obviously increase weight of unmatured rats uterus and ovaries.We concluded that we have got a highly effective way to produce anti inhibin antibody through hen.更多还原
