I1受体在家兔眼压调节中的作用

【摘要】 目的 探讨咪唑啉受体 1型 (I1 receptor)激动剂莫索尼定 (moxonidine,MOX)溶液滴眼对家兔眼压的影响 ,以及 I1 受体阻断剂 efaroxan和颈上交感神经节切除术 (superi-or cervical ganglionectomy,SCG)预处理对其效应的影响。方法 观察 7.5、2 5、15 0μg 3种不同剂量的 MOX单侧滴眼后 8h内家兔双眼眼压改变 ,并用 efaroxan滴眼和 SCG的方法分别预处理 MOX 2 5μg用药眼和对侧眼 ,观察其对 MOX滴眼眼压效应的影响。结果  3种剂量的 MOX单侧滴眼均能使家兔双侧眼压显著下降 (P<0 .1) ;efaroxan12 5 μg滴眼预处理 MOX用药眼或对侧眼 ,则 MOX2 5 μg滴眼后 8h内双眼均未观察到明显的眼压下降 ;SCG预处理可取消切除侧眼的眼压下降 ,但对非切除侧 MOX降眼压效应影响不明显。结论  MOX家兔滴眼有确切的降眼压作用 ,这种降眼压作用主要是由 I1 受体介导的 ;I1 受体介导的降眼压效应依赖于眼部交感神经结构和功能的完整。

【Abstract】 Objective To study the effects of unilateral topical administration of moxonidine (MOX) ,a selective I 1 agonist on the IOP of rabbits and the influence of the pretreatment with superior cervical ganglionectomy (SCG) or topical administration of efaroxan (I 1 antagonist) on this kind of effect.Methods The alteration of IOP in 8h period after unilateral topical administration of three different doses of MOX and the influence of the pretreatment (ipsilateral or contralateral to the MOX-treated eyes respectively) with SCG or topical administration of efaroxan on the IOP change induced by unilateral topical MOX 25μg were observed.Results Unilateral topical administration of MOX (7.5,25 and 150μg) can lower bilateral IOP significantly. Pretreatment of unilateral topical efaroxan 125μg eliminated bilateral hypotension induced by MOX 25μg in regardless of which eyes (the MOX-treated or the fellow) were pretreated. Pretreatment of the MOX 25μg-treated or the fellow eyes with unilateral SCG can inhibit the ocular hypotension on the pretreated sides and have no significant influence on the unpretreated sides.Conclusion MOX applied on rabbit eyes topically has definite hypotensive effect . I 1 receptor is involved in the hypotensive action of MOX. An intact sympathetic nerve system is required for the hypotensive effect of MOX.
