无瓣海桑Sonnerati aapertala钙调蛋白基因的克隆及序列分析
Cloning and Secquencing Analysis of Calmodulin Gene from Sonneratia apertala of Mangrove Plant
【摘要】 用高等植物钙调蛋白的保守序列设计两端引物 ,以红树植物海桑属无瓣海桑Sonneratiaapertala总DNA为模板 ,扩增出无瓣海桑钙调蛋白基因 ,并以之构建重组质粒pT ACaM .经测序分析比对 ,发现无瓣海桑钙调蛋白基因全长 1418bp ,其中第 1外显子 76bp ,第 2外显子 371bp ,中间为一 946bp的内含子所隔开 .编码 148氨基酸的蛋白质 ,其编码区序列与已知的其他高等植物的钙调蛋白基因核苷酸序列同源性高达 85 %以上 .而蛋白质序列同源性达95 %以上
【Abstract】 The sequence was amplified using total DNA of Sonneratia apertala as the template.The PCR primers was designed and synthesized according to the 5′-and 3′-terminal oligo nucleotide sequences of calmodulin gene of plants in Genbank. The result of DNA sequence analysis shows that the gene is 1 418 bp length which contains 2 exons. The first one is 76 bp, the second is 731 bp. They are interrupted by a 946 bp intron. The nucleotide sequences of ORF share more than 85% homologies as compared with those of calmodulin genes of other several plants, the amino acid sequences share more than 95% homologies with it.
