应用BACTEC-960仪进行DNA疫苗保护力评价的研究

【摘要】 根据BACTEC 96 0仪分析动物试验模型主要脏器的结核分枝杆菌生长情况 ,分析结核分枝杆菌MPT6 4和ESAT6DNA疫苗预防结核病的效力。将BALB/C小鼠随机分五组 ,分别用A组生理盐水、B组载体质粒、C组卡介苗、D组重组质粒JW 4 30 3 MPT6 4DNA、E组JW4 30 3 ESAT6DNA免疫 9周后 ,用H3 7RV结核分枝杆菌强毒株攻击小鼠。观察攻击一个月和两个月后的结果。发现C ,D ,E组比B组生长天数总体向后推迟 ,生长时间明显集中 ,生长曲峰强度比B组明显减弱 ,C组肝脏中的感染率比D ,E组有所下降。D组肺脏中感染率下降明显。D ,E组鼠脾淋巴细胞转化率较C组高 ,B明显降低。结论 :与B组比 ,小鼠免疫力的短期预防有所增强 ,其中卡介苗前期效果明显 ;MPT6 4 ,ESAT6是持续效果好。

【Abstract】 To evaluate the protective efficacy of MPT64 and ESAT6 DNA vaccine from M.tuberculosis, BALB/C mice were randomly divided into 5 groups. They were immunized with the saline (group A), plasmid vector (group B), M.bovis BCG (group C), JW4303 MPT64 (group D), JW4303 ESAT6 DNA (group E), respectively, and then injected by infection of M.tuberculosis H 37 Rv. The situation on the growth of M.tuberculosis in main organs of mice was analyzed by BACTEC 960 culture system at 1 month and 2 months after infection. The results showed that the growing days of M.tuberculosis in group C, D, E were analogous and longer than that in group B. The peaks of growth curve in group C, D, E were obviously lower than that in group B. The infective rate of liver in group C was lower than those in group D, E. The infective rate of lung in group D was descended obviously. The lymphocyte transformation rate of spleen in group A,B were lower, and those in group D,E were higher than that in group C. Therefore, the protective efficacy of M. bovis BCG was apparent in the early stage of infection, while those of MPT64 and ESAT6 DNA were strong in the middle and latter stages.
