【摘要】 目的研究软骨细胞体外培养不同时间去分化现象发生过程及规律。方法倒置显微镜观察体外培养软骨细胞形态变化;根据每代每日细胞计数结果,绘制生长曲线,检测细胞增殖能力;采用免疫组织化学方法检测软骨细胞分泌II型胶原的变化规律。结果随着培养时间的延长,软骨细胞形态发生明显的改变;细胞增殖能力及分泌细胞外基质II型胶原的能力逐渐减低。结论兔关节软骨细胞体外培养时第3代开始出现去分化现象,至第5代(8周)时去分化现象明显。更多还原

【Abstract】 Objective To investigate the dedifferentiation process and rules of articular chondrocyte in vitro.Methods Observe the change of chondrocyte shape by micro scope.According the results of cell counting every day,portray the grow th curve and detect the ability of cell proliferation.The change of Co llagen type II expression was determ inated by using immunohistochemistry.Results With the culture time Prolonged,the shape of articular ch ondrocyte change a lot,and the ability of cell proliferation and synthesis of Collagen II de-clined.Conclusions The dedifferentiation phenomena ta kes place in the 3th passage of rabbit articular chondrocyte,and it dedif ferentiates obviously in the 5th passage(8weeks).更多还原
