永久性脑缺血神经干细胞迁移及nestin蛋白表达的变化
The change in migration of neural stem cells and expression of nestin protein under the condition of permanent cerebral ischemia
【摘要】 目的进一步认识神经干细胞 (NSC)对脑损伤的反应性。方法以永久性大鼠脑缺血为模型 ,采用免疫组化染色方法 ,观察脑缺血 1,3,7,14和 2 8d时 ,NSC及其标记物nestin蛋白的变化。结果室管膜下区 (SEZ)的NSC在缺血后沿胼胝体腹侧向缺血区方向发生了迁移。其中 ,缺血 7d时达到最远。缺血灶附近在缺血 1d时 ,就出现较多的nes tin+细胞 ,缺血 3d以后逐渐减少。结论NSC对缺血性脑损伤有一定的反应能力 ,nestin蛋白表达于缺血附近可能是一种对损伤的保护机制
【Abstract】 AimTo understand further reactivity of neural stem cell(NSC) to cerebral injury. MethodsUsing rat’s permanent cerebral ischemia as a model, immunohistochemical staining were used to observe the changes in nestin protein and the migration of NSCs 1, 3, 7, 14 and 28 days after cerebral ischemia. ResultsNSCs from subependymal zone(SEZ) towards ischemic zone along ventral side of corpus callosum, the farthest migration occurred to 7 days after cerebral ischemia. In the neighbourhood of ischemic foci, more nestin + cells were observed 1 day after cerebral ischemia, and nestin + cells gradually decreased 3 day after cerebral ischemia. ConclusionThe NSCs has a certain ability to react to ischemic cerbral injury. Expressin of nestin protein around ischemic region may be a mechanism of protection from cerebral injury.
