p16和Rb在宫颈癌中的表达及其意义
Expression of p16 and Rb in cervical carcinoma:their relationship with clinicopathologic characteristics
【摘要】 目的 探讨 p1 6和Rb基因产物在宫颈癌中的表达 ,及与临床病理参数间的相关性。方法 应用免疫组织化学方法 (SP法 ) ,检测p1 6、Rb蛋白在 1 0例正常宫颈组织 ,1 0例宫颈上皮内瘤样变 (CIN)及 5 9例宫颈鳞癌中的表达。结果 2 0 %CIN (2 /1 0 )和 2 8.8%宫颈鳞癌 (1 7/5 9)中 ,p1 6蛋白表达缺失。p1 6蛋白阴性表达与病人年龄负相关 ,而与肿瘤生长类型、临床分期及组织学分级无相关性。宫颈癌中Rb表达阳性率为 88.1 4% (5 2 /5 9) ,Rb蛋白表达与宫颈癌的组织学分级呈正相关。 6 6 .1 %的宫颈癌两种蛋白共表达阳性。结论 p1 6失活参与部分宫颈癌的发生。Rb可能主要通过功能性失活参与宫颈癌的发生。宫颈癌中p1 6和Rb蛋白的表达之间不存在负相关关系
【Abstract】 Objective To study the expression of p16 and Rb in cervical carcinoma and its relationship with clinicopathologic features.Methods By using SP immunohistochemical technique, we examined the expression of p16 and Rb in 10 normal cervical tissues, 10 cervical intraepithelial neoplasias (CIN) and 59 invasive squamous cell carcinomas.Results In 20% of CIN (2/10) and 28.8% of cervical carcinomas (17/59) negative staining was shown for p16. Abnormal p16 expression was related negatively the patient’age. There were no statistically significant differences in the p16 status with respect to gross type, histologic differentiation and stage of the disease. Rb was expressed in 52 of 59 (88.14%) cases of cervical carcinomas. Rb expression was significantly correlated with histologic differentiation ( P <0.005).66.1% of the specimens were immunoreactive for both Rb and p16.Conclusion Inactivation of p16 may play an important role in the genesis of human cervical carcinoma. Rb may be functionally inactivated in cervical cancer. There seems to be no negative correlation between Rb inactivation and p16 expression in cervical cancer.
