四种抗癌物质对微管蛋白体外聚合及热变性的影响(英文)
Studies of the Effects of Anti-tumor Compounds on the Assembly and Melting Denaturation of Tubulin in vitro
【摘要】 用浊度法和差示扫描量热法研究了四种小分子抗癌物质犤紫杉醇、卡铂、猪芽藻提取物(ZYZ)和番荔枝素提取物(F43)犦对微管蛋白体外聚合及热变性的影响。实验结果表明:当四种化合物的浓度均为15μmol·L-1时,紫杉醇和猪芽藻提取物(ZYZ)能促进微管蛋白的聚合并使其热变性温度升高,而卡铂和番荔枝素提取物(F43)却对微管蛋白的聚合及热变性温度无明显影响。综合上述实验结果,对四种抗癌物质与微管蛋白相互作用的机理进行了讨论。
【Abstract】 The effects of four kinds of anti-tum or compounds,taxol,carboplat i n,Z YZ(the extractive from gleditsi-a),F43(the extractive from squamocin ),on the assembly and melting denatur ation of tubulin in vitro were studied b y turbidity and differential scannin g calorimetry.The experimental results sho w that taxol and ZYZ enhance t he assembly of tubulin,while carboplatin and F4 3have no effect on the assembly of tubulin when the concentrations of all f our kinds of compounds are 15ìmol 稬-1 .At the same time,taxol and ZYZ can ma ke the melting points of tubuin ris e,while the melting points of tubuin don’ t change in the presence of carboplat in and F43.The different interactio n mechanisms of the compounds and tubuli n have been d iscussed according to the above experimental results.
