生长抑素对胰腺癌生长抑制和凋亡作用的实验研究
Inhibition of H uman Pancreati c Cancer Growth by Stilamin in vitro and in vivo.
【摘要】 目的 :探讨外源性生长抑素类似物施他宁对胰腺癌细胞的生长抑制和凋亡作用。方法 :采用MTT和细胞流式仪观测施他宁对胰腺癌细胞的生长抑制和凋亡的作用 ,并分析其对胰腺癌移植瘤的生长影响。结果 :发现PC - 3细胞在高浓度施他宁的作用下生长缓慢 ,与低浓度比较抑制率有明显差异 (P <0 .0 5) ,并与施他宁浓度呈依赖关系。施他宁对胰腺癌细胞凋亡有促进作用 ,凋亡率为 6.31 % ,而非治疗组的细胞凋亡率为 0 .48% ,两组间有非常显著差异(P <0 .0 5)。在移植瘤中 ,施他宁治疗组在肿瘤重量和肿瘤生长速度上均小于对照组 ,其差异有统计学意义 (P <0 .0 5)。结论 :施他宁对胰腺癌生长起抑制作用和促进凋亡作用。有潜在的临床应用价值
【Abstract】 To study the effects of Somatostatin analogue Stilamin on the growth of pa ncreatic cancer in vitro and in vivo. Methods: An experimental model of transplanted human pancreatic cancer was establish ed by implanting PC- 3 human pancreatic cancer cells under the skin of nude mice . The proliferation r ate of PC-3 cells was determined by MTT method, and the ap optosis of PC-3 cell s was measured using flow cytometry assay in vitro. Results: Th e growth of human pancreatic cancer implanted in the nude mice proceeded in a do se-dependent manner, the cells grew much under high conc entration of Stilamin ( P<0.05). The apoptopic index was increas ed in PC-3 cells from 3 .46% to 19.62%. Conclusions: The Somato statin analogue Stilamin can induce apoptosis in pancreatic cancer by exercisin g its inhibitory effect o n the growth of human pancreatic cancer implanted in n ude mice.
