白介素8在新生鼠高氧肺损伤炎症反应中的作用

【摘要】 探讨白介素 8在高氧肺损伤炎症反应中的作用。采用支气管肺泡灌洗术获取肺泡巨噬细胞 ,经贴壁纯化后 ,随机分为 : 、空气组 , 、高氧组 , 、空气 +脂多糖 (lipopolysacchide,L PS)组 , 、高氧 +L PS组。培养 48h后 ,收集培养上清液 ,并检测上清液中白介素 8、过氧化氢的含量和乳酸脱氢酶活性。结果表明 :1培养 48h后 , 组白介素 8增加最显著 ,其次为 组、 组 , 组最低 ,各组之间均有显著性差异。 2 组较 组、 组较 组过氧化氢均明显降低 ; 组较 组、 组较 组过氧化氢则显著增加。 3 组、 组细胞形态无明显改变 , 组、 组细胞呈现生长不良状态 ,但乳酸脱氢酶活性在各组之间均无显著性差异。提示白介素 8参与高氧肺损伤炎症反应的发生

【Abstract】 The role of interleukin-8 in hyperoxia-induced inflammatory lung injury was investigated. Alveolar macrophages (AM) in bronchoalveolar lavage fluid (BALF) were gained from newborn SD rats of 5th to 10th days, followed by adherence and purification, and then randomly divided into 4 groups: Ⅰ. Air group; Ⅱ. Hyperoxia group, Ⅲ. Air plus lipopolysacchide (LPS) group; Ⅳ. Hyperoxia plus LPS group. After AM were cultured for 48 h, the supernatants were harvested. IL-8 levels, hydrogen peroxide (H 2O 2) levels and L-lactate dehydrogenase (LDH) activity in the supernatants were detected in all groups. Results showed that: (1) IL-8 was the highest in group Ⅳ, followed by group Ⅲ and Ⅱ, and the lowest in group Ⅰ. There were significant differences between each two groups (P<0.05, 0.05, 0.01 respectively); (2) Respectively compared with those in groups Ⅰand Ⅲ, H 2O 2 contents were decreased in groups Ⅱ and Ⅳ(both P<0.01); H 2O 2 was significantly was increased in groups Ⅲ and Ⅳ as respectively compared with those in groupsⅠand Ⅱ (P<0.01, 0.05, respectively); (3) Compared with groups Ⅰand Ⅲ, the other two groups which were given 100 % oxygen showed inhibition of cellular growth, but there was no significant difference in LDH activity among all groups. It was concluded that IL-8 might participate in hyperoxia-induced inflammatory lung injury.
