酵母核糖核酸的UV-Vis光谱探针反应的机理研究

【摘要】 采用UV -Vis分光光度法 ,研究在 pH7~8的缓冲溶液中 ,酵母核糖核酸 (RNA)与36甲氨基2基吩嗪盐酸盐 (ZR)相互作用;反应生成红色配合物 ,该配合物的最大吸收波长为540nm ;反应前后的吸收光谱变化明显 ,配合物的吸收峰与试剂本身比较红移90nm ;研究了体系的酸度、温度、时间等基本反应条件以及不同类型的离子对体系的影响 ,发现离子强度的改变对体系的吸光度有明显的影响。

【Abstract】 The interaction of ribonucleic acid(RNA) and 3 _ amino _ 6 _ dimethylamino _ 2 _ methylphenazine hydrochloride(ZR) was investigated by UV-Vis spectrophotometry in B-R buffer solution at pH 7.4. When RNA was added into ZR solution a red _ colour complex of RNA-ZR was formed with maximum absorption wavelength of 540 nm. A red _ shift of 90 nm for maximum absorption wavelength was observed. Factors affecting the absorbance of the reaction system such as acidity, temperature, reaction time, ionic type and strength were evaluated. It was found that ionic strength had a significant effect on the absorbancy of the system. The mechanism of the interaction of RNA and ZR was proposed.
