离子对反相高效液相色谱法测定兔血浆中氧化苦参碱含量

【摘要】 目的建立测定家兔血浆中氧化苦参碱含量的离子对反相高效液相色谱法。方法血浆样品用 6%高氯酸沉淀蛋白 ,上清液经碱化后以氯仿 正丁醇 (98∶2 ,V∶V)提取 ,分取有机相N2 吹干后以流动相溶解进样测定。色谱柱为DiamonsilTM C18柱 ;流动相为乙腈 水 (2 0∶80 ,V∶V ,含庚烷磺酸钠 5mmol/L,磷酸调pH 3 2 ) ,流速 1 0mL/min;检测波长为 2 2 0nm ,以非那西丁为内标物。结果在氧化苦参碱质量浓度为 0 5~ 1 0 0 μg/mL内线性关系良好 (r=0 9965 ) ,日内、日间RSD分别≤ 6 6%和≤1 3 8% ,平均回收率为 99 1 %。结论方法简便可行 ,适用于氧化苦参碱的血药浓度测定及药代动力学研究

【Abstract】 Objective To establish an IP RPHPLC method for the determination of the content of oxymatrine in the rabbit′s plasma.Methods After being precipitated with 6% HClO 4 and basificated by 5 mol/L NaOH,the organic phase was evaporated to dryness with the nitrogen stream and the residues were dissolved with the mobile phase.A Diamonsil TM C 18 column was used.The mobile phase consisted of acetonitrile and water(20∶80, V∶V , 5 mmol/L sodium octanesulfonate was contained and the pH was adjusted to 3 2 with phosphoric acid).The flow rate was 1 mL/min ,and the UV detector was set at 220 nm .Phenacetin was used as the internal standard.Results The linear calibration curves were obtained in the concentration range of 0 5~ 100 mg/L ( r =0 996 5 ).The within day and between day RSD were within 6 6% and 13 8%,respectively.The average recovery was 99 1%.Conclusions The method is simple,reliable and suitable for the determination of the concentration of oxymatrine in the plasma and the study of its pharmacokinetics.
