基质金属蛋白酶及其抑制因子在大肠癌侵袭和转移中的研究
A study on the roles of matrix metalloproteinases and tissue inhibitor of metalloproteinases in the invasion-metastasis of colorectal cancer
【摘要】 目的 探讨基质金属蛋白酶 (MMPs)及其抑制因子 (TIMPs)在大肠癌表达的意义。方法 大肠癌标本 5 0例 ,病理检验证实 :高分化腺癌 15例 ,低分化腺癌 15例 ,未分化癌 2 0例 ,大肠癌转移组 30例 ,未转移组 2 0例。采用免疫组织化学染色法检测。结果 显示 MMPs的成员 MMP9和 MMP2与大肠癌的浸润及转移密切关系。 MMP9和 MMP2阳性表达率 ,淋巴结转移组明显高于无淋巴结转移组 ;未分化癌和低分化腺癌高于高分化腺癌。 TIMP- 1和 TIMP- 2阳性表达率 ,淋巴结转移组明显低于无淋巴结转移组。结论 表明 MMP9和MMP2及 TIMP- 1和 TIMP- 2在大肠癌细胞突破基底膜向间质内浸润扩散及转移至淋巴结内起重要作用 ,两者参与了大肠癌的转移过程。
【Abstract】 Objective To understand the significance of expressions of matrix metalloproteinases (MMPs) and tissue inhibitor of metalloproteinases (TIMPs) in colorectal cancer.Methods Fifty colorectal cancer speciments from patients were used in the study,pathological examination had proved 15 as well differentiated adenocarcinoma,15 as poorly differentiated adenocercinoma,and 20 as undifferentiated carcinoma;metastatic colorectal cancer 30 cases,unmetastatic" 20 cases.Detections were made by immunohistochemical staining.Results A close relationship of MMP9,MMP2 (members of MMPs) with colorectal cancer′s invasion metastasis was observed.The positive expression rate of MMP9,MMP2 in the lymphatic metastasis group was obviously higher than that in the group without lymphatic metastasis;the above rate was also higher in undifferentiated cancer group or in poorly differentiated adenocarcinoma group than in well differentiated adenocarcinoma group.The positive expression rate of TIMP 1,TIMP 2 (members of TIMPs) in lymphatic metastasis group was markedly lower than that in the group without metastasis.Conclusion The study points out that MMP9,MMP2 and TIMP 1,TIMP 2 play an important role in the following process-colorectal cancer cells breaking into basement membrane to make a diffusional transfer to the lymph nodes.Both MMPs and TIMPs involve in the metastatic process of colorectal cancer.
