外源性p53基因增强卵巢癌细胞对化疗和放疗的敏感性

【摘要】 目的 探讨外源性p53基因转染对人卵巢癌细胞系的化疗、放疗敏感性的影响。方法 用分子克隆技术构建人野生型 p53基因与真核细胞表达载体 pcDNA3的重组体(pcDNA-p53),并用脂质体介导的转染技术,将其导入不表达p53的卵巢癌SKOV-3细胞中,经 G418筛选,Northern blot及Western blot鉴定后,观察p53基因转染对顺铂或X线放射作用的SKOV-3细胞的集落形成的影响。结果 成功地构建了人野生型p53基因与真核细胞表达载体的重组体。外源性 p53基因在转染细胞中有效表达,增强了顺铂或X线放射对 SKOV-3细胞集落形成的抑制作用。结论 外源性p53基因能增强卵巢癌细胞对顺铂或放射的敏感性,p53基因治疗与化疗或放疗联合作用能更大程度地杀灭肿瘤细胞。

【Abstract】 Objective To explore the influence of exogenous p53 gene transfection on the sensitivities of human ovarian cancer cells to chemotherapy and radiotherapy. Methods The recombinant plasmid pcDNA - p53 was constructed with human wild - type p53 gene and eukaryotic expression vector pcDNAS by molecular cloning technique and introduced into SKOV - 3 cells by lipofectin - mediated gene transfection. The expressed product was identified by Northern blot and Western blot, and the influences of cisplatin and X-ray irradiation on the colony formation of SKOV - 3 cells transfected with p53 gene were observed. Results The recombinant plasmid was successfully constructed, and the exogenous p53 was effectively expressed in transfected cells. The expression enhanced the suppressive effects of cisplatin and X - ray irradiation on the colony formation of SKOV - 3 cells. Conclusion Exogenous p53 gene enhanced the sensitivities of ovarian cancer cells to cisplatin and radiation significantly, and the combination of p53 gene therapy with chemotherapy or radiotherapy may be more effective in ovarian cancer.
