微碳纤电极与细胞量子化分泌的记录方法
MICRO CARBON FIBER ELECTRODE FOR RECORDING OF QUANTAL SECRETION IN SINGLE CELL
【摘要】 电生理研究细胞分泌有两种实时记录技术。膜电容法通过记录细胞膜表面积来估算刺激诱导的细胞分泌和胞吞导致的膜表面积总和变化。微碳纤电极(carbonfiberelectrode,CFE)法则可单纯记录细胞分泌。CFE记录灵敏度是当前最高水平 ,可记录单个50nm突触小泡的量子化递质释放。介绍CFE基本记录方法、原理和一些实验应用
【Abstract】 Biophysical technologies for single cell recording was described, with special focus on micro carbon fiber electrode (CFE) and its application in stimulus-secretion coupling. In contrast to membrane capacitance of patch-clamp recording, CFE has advantages in high sensitivity, real detection of release substance and no need for tight-seal patch-clamp, although it detects only a few easily oxidizable transmitters including catecholamines and serotonin. In combination with patch-clamping and Ca imaging measurement, coupling between action-potentials and quantal transmitter secretion in chromaffin cells are demonstrated as the example of CFE application.
