UV-B辐射和蒽对三角褐指藻DNA伤害的相互作用

【摘要】 运用生态毒理学和生物化学的方法研究了紫外线和多环芳烃——蒽对三角褐指藻 DNA的伤害作用。结果表明 ,蒽对三角褐指藻的生长有抑制作用 ;随着蒽浓度的增加 ,三角褐指藻 DNA损伤程度增加 ;在蒽浓度固定不变时 ,随着处理时间的延长 ,DNA的损伤程度同样提高 ;在蒽的处理过程中同时伴有紫外线的辐射处理 ,DNA的损伤程度加剧 ;蒽处理解除一段时间后 ,DNA损伤程度未明显减轻 ,而 UV-B处理解除后 ,DNA的损伤可明显恢复。说明 DNA的损伤可在一定程度上指示海洋微藻受蒽伤害的的程度。

【Abstract】 The paper reported that the interactive damage of UV B(280~320nm) radiation and polycyclic aromatic hydrocarbons(anthracene) to DNA of Phaeodactylum tricornutum was studied using ecotoxicological and biochemical methods. The results showed that anthracene could inhibit the growth of Phaeodactylum tricornutum, the relative growth rate in Phaeodactylum tricornutum gradually decreased with the increment of anthracene concentration, and the calculated 48h· EC 50 of anthracene to Phaeodactylum tricornutum was 0 0809mg/L. The DNA damage of algae cells was also analysized after exposure to different concentration of anthracene. The anthracene had obvious damage effect on the DNA of Phaeodactylum tricornutum, the DNA damage degree of algae cells exhibited promotion accompanied with increased exposure intensity of anthracene. Meanwhile the exposure time had effect on DNA damage of algae cells. If the anthracene concentration was fixed, the DNA damage also promoted as exposure was elongated. During early 2 days of exposure, the DNA damage degree of algae cells in exposure group had already notable difference from that in control group( p <0 05); and with the exposure time elongated(2 days later), this difference between exposure group and control group showed more remarlable( p <0 01). When Phaeodactylum tricortunum was treated with different concentration of anthracene and UV B(1 25J/m 2) radiation synchronously, the experiment results showed that UV B radiation could enhance DNA damage of algae cells caused by anthracene. This enhancement of DNA damage caused by UV B radiation had close relationship with anthracene concentration. The enhancement effect of 1 25J/m 2 UV B radiation on DNA damage in algae cells was not notable( p >0 05) as the anthracene concentration was less than 0 02 mg/L; on the contrary, it changed very remarkable compared with that under treatment by anthracene alone as anthracene concentration was more than 0 02 mg/L. The repair of DNA damage was not remarkable( p >0 05) after the algae, which were exposured to anthracene, were transferred to anthracene free fresh medium and cultured for 3 days, while DNA damage was obviously decreased( p <0 05) after the algae were continued to culture for 3 days when UV B radiation was removed. All these exhibited that the DNA damage could indicate the damage degree of marine microalgae resulted from anthracene exposure .
