扶正化瘀方对大鼠肝星状细胞旁分泌与自分泌活化途径的干预
Interference of "Resistance-Strengthening and Stasis-Dispersing Formula" on Activation Channel of Para-Secretion and Auto-Secretion in Hepatic Stallate Cells
【摘要】 为研究扶正化瘀方对肝星状细胞 (HSC)旁分泌与自分泌活化途径的干预 ,用含药 (由桃仁、丹参、虫草菌丝等组成的 3 19方 )血清分别添加于大鼠损伤肝库普弗细胞 (IKC)与传代HSC(MFBC)中培养 ,制备含药血清作用过的KC条件培养液 (IKCM)与MFBC条件培养液 (MCM) ;并以添加正常血清制备的条件培养液为对照 ,温育大鼠原代HSC。通过MTT法观察对HSC增殖与ELISA法观察对HSC分泌Ⅰ型胶原的影响。结果显示 ,IKCM与MCM可明显促进HSC的增殖与Ⅰ型胶原的分泌 ;扶正化瘀方则明显抑制了这一作用。提示扶正化瘀方对HSC旁分泌与自分泌活化途径有明显的抑制作用。
【Abstract】 In order to study interference of "Resistance Strengthening and Stasis Dispersing Formula" on activation channel of parasecretion and auto secretion in hepatic stallate cells(HSC),serum contained with drug(319 formula composed of Semen Persicae,Radix Salviae Miltiorrhizae,Cordyceps hyphae,etc.)was respectively added in rat injured Kupffers cells(IKC) and generation transmitted HSC(MFBC) for culture to prepare KC conditioned culture fluid(IKCM) and MFBC conditioned culture fluid(MCM) functioned by drug containing serum,with conditioned culture fluid added with normal serum as comparison,for breeding primary generation of rat HSC,to observe HSC proliferation by MTT and to observe influence on Type Ⅰ collagen of HSC secretion by ELISA.The results showed that IKCM and MCM could remarkably promote HSC proliferation and secretion of type Ⅰ collagen,but "Resistance Strengthening and Stasis Dispersing Formula" obviously inhibited this function,indicating that "Resistance Strengthening and Stasis Dispersing Formula" possesses an obvious inhibitory function on activation channel of para secretion and auto Secretion in hepatic stallate cells.
