【摘要】 目的　探讨冬凌草甲素对多药耐药细胞系K5 6 2 /A0 2耐药性的逆转作用。方法　以柔红霉素(DNR)和高三尖杉酯碱 (HHT)联合不同浓度的冬凌草甲素处理K5 6 2 /A0 2及敏感细胞系K5 6 2 /S ,以四唑盐比色法 (MTT)检测两种化疗药物的半数抑制浓度 (IC50 ) ,以流式细胞仪检测P170蛋白的表达与细胞内DNR的浓度。结果　冬凌草甲素可明显减小K5 6 2 /A0 2细胞对DNR、HHT的IC50 ,降低耐药倍数 ,下调P170蛋白的表达 ,增高细胞内DNR的浓度 ,而对K5 6 2 /S细胞无显著影响。结论　冬凌草甲素可逆转耐药细胞系K5 6 2 /A0 2的耐药性 ,是一种很有希望的抗白血病中药。更多还原

【Abstract】 Objective To exploe the reversal effects of Oridonin on the multidrug resistant cell line K562/A02. Methods K562/A02 and sensitive K562/S cells were treated with combination of oridonin and DNR or HHT. IC 50 values of the two chemotherapeutic drugs were analyzed by MTT, and the expression of P170 and DNR concentration in the cells were detected by flow cytometry.Results After treated with ordonin, IC 50 values of DNR and HHT of K562/A02 cell line were reduced very significantly, P <0.01 or 0.001, and the expression of P170 was downregulated and DNR concentration in K562/A02 cells upregulated significantly respectively. However, this compound had no effect on K562/S cells.Conclusion Oridonin has reversal effects on multidrug resistanct cell line K562/A02 which is a very promising anti leukemic traditional Chinese drug.更多还原
