【摘要】 目的　探讨排异反应时IL 6mRNA在心脏移植物局部动态表达情况及其与移植心自然生存时间的联系。方法　在移植术后第 1、3、5、7、9、11天切取移植心 ,提取总RNA ,将IL 6引物和对照引物 (β actin)在同一管中扩增 ,以两者像素值的比值均数作图 ,观察排异反应单一因素对IL 6mRNA动态表达的影响。结果　排异反应时心脏移植物局部的IL 6mRNA表达水平明显增高 ,其基因表达高峰在术后第 3天 ,在移植心自然死亡高峰之前。结论　IL 6在排异反应的发生发展中起着重要作用 ,IL 6mRNA的动态检测可望成为排异反应的早期诊断指标 ,抑制IL 6的措施也可望成为治疗排异反应的有效手段。更多还原

【Abstract】 Objective To dertermine the dynamic expression of IL 6 mRNA in heart allograft during rejection and to search for the correlation between the expression and duration of allograft survival. Methods The allografts and isografts were harvested in postoperative days 1,3,5,7,9,11. Following the total RNA prepared, the control primer target and primer were amplified at the same tube, the average ratio of the target products pixel volume to primer’s was calculated and the coordinate chart was made, hence the expression level of IL 6 mRNA was found directly. Results The levels of the IL 6 mRNA expression in allografts were much higher than those in the isografts. The peak of elevated level of the IL 6 mRNA expression was at the 3rd post transplantation day, and prior to the peak of death of the allografts.Conclusion The experiment suggests that the IL 6 may play an important role in rejection. Determination of IL 6 mRNA expression may be served as an useful immunologic monitoring protocol, and by the use of inhibitory measures on IL 6 may be useful for the prevention and treatment of rejection.更多还原
