用根癌农杆菌介导获得籼稻转基因植株

【摘要】 用携带p13W 4质粒的根癌农杆菌EHA10 5感染籼稻龙特甫品种的幼胚愈伤组织 ,经共培养并在潮霉素 2 5~ 5 0mg/L浓度条件下筛选 2~ 3次 ,获得了对潮霉素表现抗性的愈伤组织 ,其愈伤组织转化频率为 2 5 .3% ;将抗性愈伤组织转入分 3化培养基培养形成植株 ,经GUS活性测定和PCR检测 ,结果证明外源基因确已导入这些转基因植株之中

【Abstract】 The calli of immature embryoes of rice variety Longtefu (Oryza sativa ssp.indica) were infected by Agrobacterium tumefaciens strain EHA105 harboring Ti plasmid p13W4 carrying anti waxy gene. After cocultivation and two or three times resistant screening on the selection media which contained 25~50mg/L hygromycin,the calli resistant to hygromycin were acquired,and transformed frequency was 25.3%. The resistant calli were transferred to differentiation and regeneration media, and 9 transgenic plants were obtained. Assay of GUS activity and PCR assay of the transgenic plants indicated that the anti waxy gene had been integrated into the rice genome.
