影响大白菜子叶再生植株的因素

【摘要】 以大白菜 42号自交不亲和系、2 1 8号自交不亲和系、夏丰无菌苗子叶为外植体 ,研究了几种培养基成份对离体培养植株再生的影响。在无菌苗萌发培养基上添加 7.5mg/LAgNO3,可提高不定芽诱导频率。高压灭菌前后添加AgNO3,从大白菜子叶诱导不定芽频率无差异。降低培养基中NH4 NO3含量导致不定芽诱导频率降低。适当增减培养基中KNO3含量对不定芽诱导无影响 ,在不定芽诱导培养基中可以使用适量的2 ,4-D代替NAA ,但再生的不定芽明显畸形。

【Abstract】 Cotyledons of Chinese cabbage (Brassica pekinensis) aseptic seedlings were used as explants. Effect of medium ingredients on plant regeneration was studied. Adding 7.5mg/L AgNO 3 into germinating medium could result in a rise in the rate of shoot regeneration. There was no difference whether adding AgNO 3 into medium before autoclaving or after autoclaving. Reduction of NH 4NO 3 level could lead to a drop in the rate of shoot regeneration, while increasing or reducing KNO 3 concentration makes no influence on the shoot regeneration. If 2,4-D takes the place of NAA in the shoot induction medium, the rate of shoot regeneration remains unchanged, however, the shoots regenerated are obviously abnormal.
