丙丁酚稳定和消退动脉粥样硬化的实验研究

【摘要】 目的 :观察丙丁酚消退和稳定动脉粥样硬化 (AS)斑块的作用并探讨金属蛋白酶 (MMP1)及其组织抑制物 (TIMP1)的表达。方法 :在AS兔 ,给予丙丁酚治疗 12周。对动脉粥样硬化血管进行病理学及免疫组织化学检查 ,用RT PCR测定组织胶原I、MMP1和TIMP1的mRNA表达。结果 :丙丁酚可显著降低血脂 ,显著消退AS斑块 ;治疗后的胶原I、MMP1和TIMP1的mRNA表达水平均下降 ,其中MMP1的表达下降有统计学意义。结论 :丙丁酚有消退AS斑块的作用 ,通过减少MMP1表达 ,相对减小MMP1 TIMP1的表达比例 ,起到稳定斑块的作用。

【Abstract】 Objective:To study the effect of probucal on regression and stabilization of atherosclerotic plaque, and the expression of matrix metalloproteinase(MMP1) and its tissue inhibitor(TIMP1). Methods:Twenty five rabbits were induced to develop atherosclerosis by cholesterol feeding and then treated with probucal for 12 weeks. Pathology and immunohistochemistry staining were employed in the study. The mRNA level of collagen 1, MMP1, and TIMP1 were analysed by RT PCR.Results:Probucal could reduce blood lipids and regress atherosclerotic plaque significantly. The mRNA level of collagen 1,MMP1 and TIMP1 were decreased in probucal group.Conclusion:Probucal can regress atherosclerotic plaque of the rabbits and can also stabilize the plaque by reducing the expression of MMP1 and MMP1/TIMP1.
