补阳还五汤对神经干细胞生长分化的影响

【摘要】 目的 :观察补阳还五汤对神经干细胞生长分化的影响。方法 :取孕 12d大鼠胚胎神经管上皮细胞 ,分别接种在灌服补阳还五汤的鼠血清和对照鼠血清的培养基内。不同培养时间内观察细胞的生长状况 ;用MTT法检测细胞活性 ;用免疫组化染色法检测神经元特异烯醇化酶 (NSE)和胶质纤维酸性蛋白(GFAP)的表达。结果 :药物血清组神经干细胞的生长明显好于对照组 ,OD值高于对照组 ;免疫组化染色显示 ,NSE、GFAP免疫阳性细胞药物血清组均多于对照组。结论 :补阳还五汤药物血清可促进神经干细胞的生长、存活和向神经元及神经胶质细胞的分化。

【Abstract】 Objective:To evaluate the action of buyanghuanwu decoction(BYHWD) on the growth and differentiation of neural stem cells in culture.Methods:Neuroepithelial cells were dissociated from neural tube of embryonic E12 rats and cultured in the serum of rats given BYHWD and the serum of normal controls. Micromeasure technique was used to detect the length of the longest process, MTT assay was applied to reveal the OD value,and immunohistochemical assay was used to identify the expression of neuro specific enolase (NSE) and glial fibrillary acidic protein (GFAP) in cultured cells.Results: The serum of rats fed with BYHWD promoted the growth of process and elevated OD value compared with the control group. Immunohistochemical staining showed that more NSE positive and GFAP positive cells were found in the experimental groups.Conclusion:The serum of rats fed with BYHWD can promote the growth and differentiation of neural stem cells.
