氧化还原电解脱保护DNA在片合成新方法的探讨
In Situ Synthesis of DNA Microarray:Deprotectionwith Acid by Electrolysis
【摘要】 DNA芯片是一种新的高通量DNA分析检测技术。DNA芯片把大量已知序列探针集成在基片上 ,通过与标记的若干靶核苷酸序列杂交 ,可以对生物细胞或组织中大量的基因信息进行检测和分析。DNA芯片的制备方法大致可以分为点样法和在片合成法。本文初步探索了以氧化还原电解脱保护法为特征的电助基因芯片制备方法 ,获得了一些有意义的结果
【Abstract】 DNA Chip technology has attracted many anentions in recent years.DNA chips or DNA microarrays,anchor tens of thousands of closely arrayed oligonucleotide probes on the surfaces of solid substrates. Through the process of base matching,every oligonucleotide sequence on a chip can recognize its complementary sequence, so this highly parallel recognition can give abundant biologic information. Most of the low-density DNA chips are prepared by spotting, and high-density DNA chips in situ synthesis. This paper reported a novel approach to synthesize DNA microarrays in situ by means of deprotection by electrolysis.
