抗除草剂Bar基因对小麦的遗传转化
Genetic Transformation by Transferring Herbicide Resistance Bar Gene into Wheat
【摘要】 作者通过根癌农杆菌 (Agrobacteriumtumefaciens)Ti质粒介导和基因枪法 ,将含抗除草剂Bar基因的植物表达载体 ,导入小麦幼穗和幼胚愈伤组织 .X Gluc染色检查表达载体中GUS标记基因的瞬时表达 ,并在含有除草剂Basta的培养基上进行多步筛选 .以愈伤组织为单位 ,统计GUS表达阳性频率和转化体的分化频率 .结果表明 :基因枪轰击法的平均转化率高于农杆菌导入法 ;幼穗作为受体的转化率高于幼胚 ;干燥处理有利于提高根癌农杆菌对小麦愈伤受体的浸染力 .
【Abstract】 The plant expression vector carrying a herbicide resistance BAR gene and a marker GUS gene was transferred into wheat spikelets and immature embryo calli via Agrobacterium tumefaciens Ti plasmid and microprojectile bombardment. The activity of GUS gene expression products was detected by X-Gluc histochemical dying and the resistance calli were selected on Basta containing medium. The GUS expression rate and regeneration rate were calculated to get the transformation frequency. The results showed that the frequency of transformation via microprojectile bombardment is higher than that of Agrobacterium tumefaciens; the frequency of transformation into spikelets is higher than that of immature embryo calli; drying was favorable for Agrobacterium tumefaciens to influence wheat receptor.
