盾叶薯蓣组织培养与快速繁殖研究
Studies on Tissue Culture and Rapid Propagation of Dioscorea Zingiberensis
【摘要】 对盾叶薯蓣的茎、叶片、叶柄进行了愈伤组织的诱导、分化 ,以及多芽体的诱导、生根、移栽的初步研究 .结果表明 :(1)茎的愈伤组织诱导率最高 ,在MS + 6 BA 0 .5mg/L + 2 ,4 D 2 .0mg/L上可达 87.5 % ,但愈伤组织不易分化出苗 ;(2 )腋芽能萌发形成多芽体 ,月繁殖系数达3~ 5 ,最适宜的多芽体诱导培养基是MS + 6 BA 2 .0mg/L +NAA 0 .5 1.0mg/L +椰汁 10 % ;(3)无根苗在MS + 6 BA 2 .0mg/L +NAA 2 .0mg/L培养基上能形成完整的再生植株 .
【Abstract】 Preliminary studies were done on callus induction and differentiation, multi buds induction, root formation and transplantation of Dioscorea zingiberensis. The results showed: (1) The callus induction rate of stem was the highest(87.5%)on MS+6 BA 0.5mg/L+2,4 D 2.0mg/L medium, but callus was difficult to differentiate buds. (2) Multi bud bodies could be induced from axillary buds, the appreciate medium was MS+6 BA 2.0mg/L +NAA 0.5~1.0mg/L+coconut milk10% and the monthly propagation ratio was 3~5. (3) Complete regenerated plantlets could be obtained on MS+6 BA 2.0mg/L+NAA 2.0mg/L medium.
