用RAPD技术分析出血性大肠杆菌O157∶H7
DNA Polymorphism of Enterohemorrhagic Escherichia coli O157∶H7
【摘要】 目的 应用RAPD技术分析肠出血性大肠杆菌O1 57∶H7DNA多态性并对其分型。方法 用两条1 0bp的随机引物对 3株肠出血性大肠杆菌O1 57∶H7和 3株其它病原性大肠艾希氏菌进行扩增 ,扩增产物经凝胶电泳后得到指纹图。结果 共得到 1 2个不同的DNA指纹图谱 ,经统计软件SPSS系统聚类分析得到两个不同的分类树状图。结论 RAPD技术方法简便、快速、分辨力高 ,在肠出血性大肠杆菌O1 57:H7分子流行病学研究中有较大的应用前景
【Abstract】 Objective To study the DNA polymorphism and identify the subtypes of E.coli O157∶H7 by using Randomly Amplified Polymorphic DNA. Methods Two groups of 10bp oligonucleotide primers were used for the RAPD analysis of 3 strains of enterohemorrhagic E.coli O157∶H7 and 3 strains of other E.coli. Result Altogether 12 DNA fingerprints and two different dendrograms were obtained from the RAPD analysis and by SPSS, respectively. Conclusion RAPD was a simple and quick method in molecular epidemiological studies to distinguish different strains of enterohemorrhagic E.coli O157∶H7.
