中华硬蜱叮咬105kDa纯化抗原免疫接种宿主后中肠组织化学变化的动态观察

【摘要】 目的 观察中华硬蜱叮咬 10 5kDa纯化抗原免疫接种宿主后 ,其中肠组织化学变化。方法 采用茚三酮法、PAS法、Feulgen法及图像分析仪 ,对中华硬蜱叮咬不同免疫力宿主后中肠蛋白质、糖原及多糖定位和分布以及DNA的含量进行研究。结果 中华硬蜱叮咬 10 5kDa纯化抗原组宿主后 ,中肠上皮细胞蛋白质含量较对照组明显降低 ,但糖原及多糖含量与初次叮咬或佐剂对照组相比无显著性变化 ;随着叮咬时间的延长 ,纯化抗原组DNA含量明显低于对照组 ,且以 2~ 4倍体为主。结论  10 5kDa纯化抗原组诱导的抗蜱免疫作用可干扰中肠消化细胞蛋白质和核酸的合成 ,而对中肠糖代谢无明显影响

【Abstract】 Objective To observe the dynamic changes of histochemistry in the midgut of ticks (Ixodes sinensis) after infesting on rabbits immunized with 105kDa purified antigen. Methods The techniques of protein staining, PAS, Feulgen method and imaging analysis were employed to determine the content of protein, glycogen, polysaccharide and DNA in the digestive cells of ticks’ midgut.Results The results showed that the content of protein in the digestive cells of the ticks infesting on immunized hosts was obviously lower than that of ticks first infesting on hosts. But the content of glycogen and polysaccharide showed no difference between the ticks infesting on the immunized hosts or un-immunized host or the ticks of first infesting on host. DNA content of digestive cells doubled at 24h post infesting. With the increasing time of hematophagia, the enlargement of the nucleus volume was up to 7.06 times on the 5th day. But the increase of the nucleus volume was not obvious when the ticks infested on immunized hosts. Conclusion The results indicated that after infesting the host immunized with purified antigens, the synthetic metabolism of the protein and DNA in the ticks’ digestive cells of midgut might be affected, but no effect on polysaccharide metabolism.
