血管内放射对颈动脉内膜剥脱术后NO和ET水平的影响

【摘要】 目的 :通过观察血管内放射对颈动脉内膜剥脱术 (CEA)后NO和内皮素 (ET)水平变化 ,探讨内放射治疗防治再狭窄的可能作用机理。方法 :2 4只兔颈动脉内膜剥脱术后 ,随机分为 3组 ,分别给予 0、10、2 0Gy放射剂量 (32 P) ,术前 3d、术后 3、7、14、2 8d耳缘静脉各抽血 4ml,分别测量NO、ET 1浓度 ,观察其水平变化。结果 :术后NO水平内放射组较对照组明显升高 (P <0 .0 1) ,而ET 1水平明显降低 (P <0 .0 5 )。 10Gy、2 0Gy之间相比未见显著差异。结论 :内放射治疗可能是通过增加NO、降低ET 1含量来抑制平滑肌细胞增生、迁移 ,防治RS ;血浆NO、ET 1水平的变化可作为一种观察疗效的指标

【Abstract】 Objective:To find the possible mechanisms of endovascular radiation in the prevention of restenosis in rabbits following carotid endarterectomy and to observe the dose effect relationship.Methods:Twenty four rabbits were randomly divided into three groups following carotid endarterectomy (each n=8) and were allocated to receive a radiation dose of 0,10,20Gy( 32 P)respectively.Changes in the levels of plasma nitric oxide(NO) and endothelin 1(ET 1)were measured 3 day before operation and 3,7,14,28 day after operation.Results:The plasma NO was markedly increased and ET 1 was markedly decreased in endovascular radiation groups compared to control groups.The results were similar in the two groups(10Gy versus 20Gy).Conclusion:The endovascular radiation may inhibit smooth muscle cell proliferation and migration,and thus prevent restenosis by increasing NO and decreasing the level of ET 1.Changes of the levels of plasma NO and ET 1 is a practicable methods to detect the effect of therapy.
