家蚕蛹表达乙肝表面抗原中蛋白免疫原性的研究
Immunizing Effect of HBV Surface Antigen Middle Protein Expressed in Silkworm Pupae
【摘要】 将含有HBV(adr)的表面抗原中蛋白基因的重组病毒。BmPAK-HBM感染家蚕(Bombyx mori)蛹,ELISA和Westernblotting分析表明,家蚕蛹表达产物具有较高的HBsAg抗原性,并能检测到PreS2抗原性。将重组病毒感染蛹表达的乙肝表面抗原中蛋白对SD鼠进行注射免疫和口服免疫,注射免疫后的鼠体内能产生较强的特异性抗-HBs抗体和抗-PreS2抗体;口服免疫实验表明,连续添食6周后,约40%的鼠体内产生了免疫应答反应,在抗体转阳的鼠体内能同时检测到抗-HBs和抗-PreS2抗体。
【Abstract】 The HBsAg and PreS2-Ag were detected in silkworm (Bombyx mori) pupae infected with recombinant virus BmPAK-HBM (containing HBV surface antigen middle protein gene) by ELISA. HBsAg expressed in silkworm pupae was analyzed by SDS-PAGE and immunoblotting with an anti-HBs antibody, and four bands were detected by Western blotting corresponding to molecular weights of 25(P25), 28(GP28), 30(GP30) and 31(P31) kD. Further analysis showed that the products expressed in pupae elicited stronger antibody response against S and PreS2 antigens in SD mice by injection immunization, and 40 % mice elicited antibody response against S and PreS2 antigens by oral immunization.
