柚(Citrus grandis Osbeck)第一染色体的显微分离
Microdissection of the First Chromosome in Pomelo(Citrus grandis Osbeck)
【摘要】 本研究利用显微操作技术分离木本果树植物柚单个染色体。以琯溪蜜柚的20个良好的染色体分裂相为基础,建立琯溪蜜柚染色体核型图,并以此为依据,识别第一染色体。经显微操作,将单条第一染色体放入Eppendorf管中,为木本植物小型染色体的微切割、微分离和微克隆奠定了基础。
【Abstract】 Single chromosome of pomelo (Citrus grandis Osbeck)were successfully isolated with glass needles by means of micromanipulation.This is the first time that the micromanipulation technology was applied to establish a system of single chromosome microdissection in woody fruit trees.The standard karyotype of pomelo (Citrus grandis cv.Guanxi)was established based on 20 early metaphase photos.The first chromosome was identified according to the standard karyotype of the pomelo,and then microdissected and transferred into Eppendorf.The pomelo chromosomes are very small at the metaphase,with an average of only about 1μm in length.Therefore,this research provides a plausible chance for microdissection and microcloning of single tiny chromosome in woody plants.
