【摘要】 目的　以高表达CEA的人结肠癌细胞系LoVo作为抗原 ,建立检测抗CEA抗体的细胞 ELISA法。方法　将1.0× 10 6 mL浓度 2 0 0 μL的LoVo加入培养板培养 2 4h ,用 0 .0 2 5 %戊二醛固定 ,加入经CEA基因疫苗免疫小鼠血清 ,亲和素 生物素复合物 ,酶标仪检测OD值。结果　建立了ABC CELISA检测抗CEA抗体 ,2套基因疫苗诱导抗CEA抗体效价不同。结论　ABC CELISA法简便 ,敏感度高 ,为研究CEA体液免疫提供了方法。更多还原

【Abstract】 Objective To construct Cell ELISA method and detect CEA antibody with human colonic LoVo cell line which expresses CEA highly. Methods Coat wells of cell culture plate with 200 μL of 1.0×10 6/mL LoVo cell for 24 h. Fix with 0.025% glutaraldehyde, then add sera of mice which were immunized by CEA gene vaccines, avidin biotin complex. Monitor the OD values by spectrophotometer. Results We successfully established ABC CELISA method for detection of anti CEA antibody. Two sets of gene vaccines induced anti CEA antibody level differently. Conclusion ABC CELISA method is simple, sensitive and can be used in the study of CEA related humoral immunity.更多还原
