小麦辐射雄性不育系89AR及其保持系和恢复系线粒体DNA的RAPD分析
RAPD Analysis on Mitochondrial DNA of the Cytoplasmic Male Sterile Lines 89AR Induced by Radiation and Its Maintainer and Restorer
【摘要】 8 9AR是通过辐射结合远缘杂交获得的新疆小麦亚种细胞质雄性不育系。采用 RAPD技术对 89AR及其保持系原冬 3号和恢复系 BPM1 5的线粒体 DNA(m t DNA)进行了比较研究。结果表明 ,89AR与其保持系原冬 3号之间扩增带的差异率为 38.9% ,而原冬 3号和 BPM1 5之间差异仅为 14 .9% ,不育系与保持系 m t DNA之间的差异显著 ,推测这种差异与不育性有关。为了进一步研究 89AR的雄性不育机理 ,我们采用 RAPD技术分析了同质异核的不育系 89AR(原冬 3号 )与 89AR(原冬 3号 )× BPM1 5杂种 F1 的 m t DNA的多态性扩增 ,结果表明 ,不育系与其杂种F1 之间即同质异核间 mt DNA扩增带存在差异 ,这表明细胞核通过与细胞质的相互作用而使其表达为可育或不育
【Abstract】 AR was a cytoplasmic male sterile line which was obtained from Xinjiang wheat distant cross with pollens of Leymus racemosus irradiated by gamma ray. A comparative analysis for mitochondrial DNAs(mtDNAs) between 89AR and its maintainer and restorer were performed by using RAPD techniques. The results showed that there was significant difference between 89AR(Yuandong3) and Yuandong3. It suggested that the constructural difference on mtDNA were related to male sterility. Further study results showed that the express of fertility was controlled by interaction between cytoplasm and nuclear.
