小麦耐盐突变体生化标记的研究
Study on Biochemical Markers of the Wheat Salt-tolerant Mutants
【摘要】 以两个普通小麦耐盐突变体 890 1 17和 974 915及其各自的亲本为材料 ,依据前人的工作选择第一、三、四、六同源群染色体上的生化标记进行了耐盐突变体突变位点的定位研究。高分子量麦谷蛋白亚基 (HMW GS)SDS PAGE ,EST 5、β Amy 1、α Amy 1等电聚焦 (IEF)电泳分析结果显示 :974 915与其父母本在HMW GS、βAmy 1谱带上表现出了明显的差异带型 ;890 1 17与其亲本冀麦 2 4在α Amy 1谱带上表现两条与耐盐性有密切关系的差异带。两个突变体与其相应亲本在EST 5上均未表现出差异。由此可见 ,突变体 974 915的突变位点主要发生在第一、四同源群上 ,而 890 1 17的突变位点则主要发生在第六同源群上。该结果为RFLP定位耐盐基因时选择合适的探针提供了依据
【Abstract】 and 974915, two wheat salt tolerant mutants, and their respective parents were used in this study . On the basis of the previous work, the location of the mutation sites of salt tolerant mutants were studied by the biochemical markers of HMW GS, EST 5, β Amy 1 and α Amy 1 in this article. SDS PAGE separation of HMW GS and isoelectric focusing (IEF) separation of β Amy 1 showed out differences among 974915 and its parents, Bainong 3039 and Punong 3665. In the pattern of α Amy 1, two special bands showed close relation to the difference of salt tolerant ability between Jimai24 and its mutant, 8901 17. No variations were observed in the pattern of EST 5 between the two mutants and their respective parents. These results implied that the mutation sites of 974915 most probably happened on the first and the fourth homologous group chromosomes, and that of 8901 17 on the sixth homologous group. These will be helpful to choose proper probes of RFLP molecular marker of the salt tolerant genes in the future work.
