大鼠局灶性脑缺血再灌注后转化生长因子β1表达的研究
The expression of TGF-β1 protein in rat after focal cerebral ischemia and reperfusion
【摘要】 目的 观察大鼠大脑中动脉阻塞再灌注 (MCAO R)后转化生长因子β1(TGF β1)表达的变化及其意义。方法 采用免疫组织化学方法检测TGF β1在缺血再灌注脑组织损伤中的变化情况。 结果 再灌注后 6h ,缺血再灌注组脑组织TGF β1阳性细胞数较相应的假手术组和正常对照组显著增加(P<0 .0 1) ;正常组、假手术组仅在少数血管中有微弱的TGF β1阳性表达 ,而缺血再灌注组脑梗死区神经元、胶质细胞和血管内皮细胞和平滑肌细胞中TGF β1均有显著增强的阳性表达 ,另外 ,在软脑膜血管内皮细胞和平滑肌细胞中也可见TGF β1阳性细胞表达。 结论 脑缺血再灌注损伤可诱导TGF β1表达持续增高
【Abstract】 Objective To study the expression of transforming growth factor beta1 (TGF β1) at different time points of reperfusion after middle cerebral artery occlusion (MCAO).Methods The expression of TGF β1 protein was measured by immunohistochemical technology.Results From the sixth hour of reperfusion, the TGF β1 positive cells at ischemic area in operation groups increased as compared with those in sham operation and control groups (P<0.01).Only a few vascular endothelial cells showed a weak labeling after staining with TGF β1 antiserum in normal controls and sham operated controls. In MCAO group,the most marked up regulation occurred in vascular endothelial cells and smooth muscle cells of the infarct and the border zones. Neurons and gliacytes in the border zones showed a moderately intense expression. Occasionally some leptomeningeal and cortical vascular endothelial cells and smooth muscle cells showed a positive staining reaction.Conclusion High level of TGF β1 expression persists for a long time following ischemia reperfusion injury of brain and is an endogenous self protective mechanism,which helps to maintain survival of neurons and recover neural function.
