薄层层析法测定纤维素材料水解液中混合糖的研究

【摘要】 提出了用薄层层析测定纤维素材料水解液混合糖中单糖和各种多糖的含量 ,以硅胶G为固体吸附剂 ,以玻璃为支撑物 ,制成薄层板。以V正丁醇∶V37%乙酸∶V水 =3∶2∶2为展开剂 ,以苯胺 草酸为显色剂 ,分离效果良好 ,可完成定性和定量分析任务

【Abstract】 A study on the TLC method for the determination of monosaccharose and polysaccharose in the hydrolytic solution of plant cellulose is reported. The TLC plate is prepared by applying the adsorbing material, silica gel G in colloidal solution, on to the glass plate of 20cm×5cm in dimension. A mixed solution composing of n butyl alcohol, 37% acetic acid and water in a ratio of V n C 4H 9OH ∶ V 37CH 3COOH ∶ V H 2O =3∶3∶2 by volume is used as development agent. A solution containing aniline and oxalic acid dissolved in water saturated n butyl alcohol is used as color reagent. Satisfactory separation is acquired as shown by the thin layer chromatogram. Qualitative results can be obtained in accordance to the R f value of the standard spots. Quantitative analysis is performed by photometry after coloration.
