板蓝根高级不饱和脂肪组酸的体外抗人肝癌BEL-7402细胞活性

【摘要】 目的 :研究板蓝根高级不饱和脂肪组酸体外抗人肝癌BEL 74 0 2细胞的活性。方法 :MTT法测定细胞毒作用 ,集落形成实验观察药物对肝癌细胞增殖的抑制作用 ,PCR ELISA试剂盒测定细胞的端粒酶活性。结果 :板蓝根高级不饱和脂肪组酸能抑制BEL 74 0 2细胞的增殖 ,其IC50 为 0 6mg·mL-1,有促使肿瘤细胞向正常细胞逆转的趋势 ,可降低端粒酶活性的表达。结论 :板蓝根高级不饱和脂肪组酸具有体外抗肿瘤活性

【Abstract】 AIM: To study the anti cancer activity of high grade unsaturated fatty acids from Isatis tinctoria L on BEL 7402 human hepatocellular carcinoma cell in vitro . METHODS: Cytotoxicity assay was tested by MTT method. The proliferation of BEL 7402 cells was observed by colony forming units assay. Telomerase activity in cell was determined by PCR ELISA method. RESULTS: IC 50 of unsaturated fatty acids from Isatis tinctoria L on BEL 7402 cell line was 0 6 mg·mL -1 . Telomerase activity was dramatically declined during the cell incubation with unsaturated fatty acids. Unsaturated fatty acids can induce the BEL 7402 cells to reverse normal cells. CONCLUSION: Unsaturated fatty acids from Insatis tinctoria L possesses anti tumor activity in vitro .
