TGF-β1和TIL在喉癌局部免疫反应中作用的研究

【摘要】 目的 :检测喉癌组织TGF β1的表达 ,分析TGF β1与喉癌临床指标的关系 ,明确TGF β1表达对TIL细胞浸润和局部免疫功能的影响。方法 :38例原发性喉癌标本 ,应用免疫组化SP法 ,检测喉癌组织CD3 、CD4 、CD8、CD57、CD68和CD2 5的浸润及TGF β1的表达 ;同时以 10例声带息肉做对照组。结果 :与对照组相比 ,喉癌组织TGF β1蛋白表达减少 ,且TGF β1表达与喉癌病人性别、年龄、临床分期及不同发生部位无关 ,随着恶性度的增高 ,TGF β1表达减少 ,淋巴结转移组TGF β1阳性率多于未转移组 ,TGF β1表达与CD4 细胞浸润无关 ,而与CD8浸润有显著正相关 ,TGF β1表达能诱发CD4/CD8比值倒置。结论 :喉癌组织TGF β1蛋白表达减少 ,TGF β1对喉癌细胞的生长可能起抑制作用 ,有利于肿瘤细胞的转移 ,通过影响CD8细胞的浸润 ,改变CD4/CD8比值 ,进而抑制肿瘤局部免疫功能。

【Abstract】 Objective:To study the expression of TGF-β 1 in laryngeal carcinoma and its relationship to clinical indicator, then determine the effect of TGF-β 1 to TIL cell and local immune function.Methods:CD 3?CD 4?CD 8?CD 57?CD 68?CD 25 and TGF-β 1 were detected in thirty-eight samples of primary laryngeal carcinoma by SP immunohistochemistry, and ten samples of vocal cord polypus were set as control.Results:compared with control group, decreased expression of TGF-β 1 in laryngeal carcinoma has no relation to sex, age, clinical stages and different sites. Along with the malignant degrees elevating, TGF-β 1 lowers. The positive rate of TGF-β 1 in lymph node metastasis group is higher than that in lymph node unmetastasis group, TGF-β 1 which has no relation to CD 4 cell infiltration, however has postive relationship to CD 8 cell can induce CD4/CD8 ratio inversion.Conclusion:TGF-β 1 which is decreased in laryngeal carcinoma can inhibit the growth of carcinoma cell and is in favour of metastasis of carcinoma, restraining the local immune function by influencing CD 8 cell infiltration and altering the CD4/CD8 ratio.
