Fas抗体对小鼠膀胱癌的体内免疫治疗作用

【摘要】 目的 :研究Fas抗体对小鼠膀胱癌的体内免疫治疗作用。方法 :采用仓鼠抗小鼠Fas单克隆抗体RK8对小鼠可移植性膀胱移行细胞癌模型BST73 9进行体内免疫治疗。结果 :免疫组织化学方法证明小鼠可移植性膀胱移行细胞癌模型BST73 9表达Fas;将仓鼠抗小鼠Fas单克隆抗体RK82 5 μg直接注射于 1只荷瘤小鼠尾静脉内 ,导致小鼠在 13~ 15h内死亡 ,DNAladder方法证明小鼠肝脏具有明显的凋亡性特征 ;对 6只荷瘤小鼠肿瘤局部多点分别注入仓鼠抗小鼠Fas单克隆抗体RK82 0 μg后 ,荷瘤小鼠平均生存时间为 12d ,明显长于对照组的6 .33d(P <0 .0 5 )。结论 :直接局部注射Fas单克隆抗体对表达Fas的小鼠膀胱实体肿瘤具有治疗作用 ,可明显延长荷瘤小鼠的存活期 ,且无明显的全身毒副作用。

【Abstract】 Purpose:The aim of this study was to research in vivo immunotherapy effect of anti Fas antibody on mice bladder carcinoma. Methods:With hamster anti mouse Fas monoclonal antibody IgG RK8, in vivo immunotherapy of mouse bladder solid carcinoma BST 739 was performed through vein and direct local injection.Results:It was demonstrated that the tissue of mouse bladder carcinoma BST 739 express Fas protein; direct injection of 25 μg hamster anti mouse Fas monoclonal antibody IgG RK8 into tail vein of mouse with bladder carcinoma BST 739 caused the mouse death within 13~15 hour, with the feature of hepatic apoptosis demonstrated by DNA ladder; mice received multi point injection of 20 μg hamster anti mouse Fas monoclonal antibody IgG RK8 directly into each solid tumor tissue of 6 bladder carcinoma BST 739 mice had an average survival period prolonged to 12 days, significantly longer than that in control group’s 6.33 days (P< 0.05 ).Conclusions:Without significant systemic toxicity, local direct injection of Fas monoclonal antibody into solid tumor tissue had therapeutic effect on carcinoma cells expressing Fas.
