加速器质谱技术检测细胞中DNA加合物的方法体系的建立及其应用
The Assay of DNA Adducts in Cell System by AMS and Its Application
【摘要】 本研究采用加速器质谱技术对体外培养细胞进行化学致癌物——苯并芘及杀灭菊酯的DNA加合物的C的标记示踪研究,建立了DNA加合物的剂量效应关系,并初步探讨了化学致癌物与细胞作用时间与DNA加合物形成关系,建立的该细胞模型可为细胞水平的化学致癌的机理的探讨提供有力的手段。
【Abstract】 Accelerator mass spectrometry (AMS) was used to study DNA adducts of chemical carcinogens——benzo (a)pyrene and sumicidin by 14C tracing in vitro cell culture model. The relationships between adducts and exposure were established and relationships of adduct formation and exposure time have been primarily studied. This cell model can be used to study the mechanisms of chemical tnmorigenesis by carcinogens on cell level.
